Product Description

Indications:Vefend is an azole antifungal used in patients aged 12 years and above for serious fungal infections such as invasive aspergillosis, candidemia, esophageal candidiasis, and infections caused by Scedosporium and Fusarium. It is also used in patients who do not respond to or cannot tolerate other antifungal treatments.Pharmacology:Voriconazole is a triazole antifungal that inhibits fungal ergosterol synthesis by blocking CYP450-dependent enzymes, leading to disruption of fungal cell membrane and cell death.Dosage and Administration:Oral: Take at least 1 hour before or after meals.≥40 kg: Loading 400 mg every 12 hours (first 24 hours), then 200 mg twice daily.<40 kg: Loading 200 mg every 12 hours, then 100 mg twice daily.Injection:Loading: 6 mg/kg every 12 hours (first 24 hours).Maintenance: 3–4 mg/kg or 4 mg/kg every 12 hours depending on infection type.Drug Interactions:Interacts with CYP3A4, CYP2C9 and CYP2C19 inhibitors/inducers. May increase levels of many drugs. Dose adjustments and monitoring are required.Contraindications:Contraindicated in hypersensitivity to voriconazole and with drugs like terfenadine, cisapride, quinidine, rifampin, carbamazepine, ergot alkaloids and others due to serious interaction risks.Side Effects:Common side effects include abdominal pain, nausea, diarrhea, headache, anemia, chest pain and visual disturbances.Pregnancy and Lactation:Use during pregnancy only if benefits outweigh risks. Safety in breastfeeding is not well established.Precautions and Warnings:Long-term use (>180 days) requires careful monitoring. Risk of skin cancer (SCC) has been reported with prolonged therapy.Use in Special Populations:Safety and effectiveness in children under 12 years are not established.Overdose Effects:No specific data available. Symptomatic and supportive treatment is recommended.Therapeutic Class:Antifungal (Azole group)Storage Conditions:Store below 25°C in a dry place, protected from light.
