Glucosamine Sulfate and Diacerein is a synergistic fixed-dose combination primarily used as a Symptomatic Slow-Acting Drug for Osteoarthritis (SYSADOA). Glucosamine Sulfate, a naturally occurring amino sugar, serves as a fundamental building block for the synthesis of glycosaminoglycans and proteoglycans in the articular cartilage matrix, promoting repair and maintaining joint lubrication. Diacerein, an anthraquinone derivative, provides a complementary anti-inflammatory effect by inhibiting the production and activity of interleukin-1 beta (IL-1β), a key cytokine responsible for cartilage degradation and synovial inflammation. This combination is designed to not only alleviate symptoms like joint pain and stiffness but also to potentially retard the structural progression of the disease. While Glucosamine is generally well-tolerated, Diacerein may cause gastrointestinal side effects, most notably diarrhea and a harmless discoloration of urine (pseudo-hematuria). It is important to note that this combination has a slow onset of action, typically requiring 2 to 4 weeks of consistent administration to achieve noticeable clinical benefits, and it is often contraindicated in patients with significant renal impairment or active liver disease.
