Indications:
Ceftizone is used for the treatment of serious bacterial infections including lower respiratory tract infections, acute otitis media, skin and soft tissue infections, urinary tract infections, gonorrhea, septicemia, bone and joint infections and meningitis. It is also used for prevention of postoperative infections and surgical prophylaxis.
Pharmacology:
Ceftriaxone is a third-generation cephalosporin antibiotic with broad-spectrum bactericidal activity. It works by inhibiting bacterial cell wall synthesis. It is highly stable against beta-lactamases and has a long half-life (6–9 hours), allowing once-daily dosing. It is highly protein-bound and excreted through urine and bile.
Dosage and Administration:
Adults usually receive 1–2 g once daily (IV/IM), which may be increased up to 4 g/day depending on severity.
Gonorrhea: 250 mg IM single dose.
Surgical prophylaxis: 1 g IV before surgery.
Children (≥1 month): 50–75 mg/kg once daily (max 2 g/day).
Meningitis: up to 100 mg/kg/day (max 4 g/day).
Treatment duration is usually 4–14 days or as directed.
Drug Interactions:
No significant drug interactions have been reported.
Contraindications:
Contraindicated in patients with hypersensitivity to cephalosporins.
Side Effects:
Generally well tolerated. Possible effects include diarrhea, nausea, rash, itching, hematologic changes, liver enzyme elevation and rarely local injection site reactions or phlebitis.
Pregnancy and Lactation:
Use during pregnancy only if clearly necessary. Small amounts are excreted in breast milk, so caution is advised in breastfeeding.
Precautions and Warnings:
Risk of allergic reactions including anaphylaxis. Monitor blood counts during prolonged use. Gallbladder sludge may occur but is usually reversible.
Overdose Effects:
No specific antidote; treatment is symptomatic and supportive.
