Etorix, which contains etoricoxib, is an oral non-steroidal anti-inflammatory drug (NSAID) that belongs to the class of selective COX-2 inhibitors (coxibs). It is primarily used to relieve pain and reduce inflammation in conditions such as osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and acute gouty arthritis. Unlike traditional NSAIDs (like aspirin or ibuprofen) that inhibit both COX-1 and COX-2 enzymes, etoricoxib specifically targets the cyclooxygenase-2 (COX-2) enzyme responsible for producing prostaglandins that cause pain and swelling. By sparing the COX-1 enzyme, which helps protect the stomach lining, etoricoxib is generally associated with a lower risk of gastrointestinal ulcers and bleeding compared to non-selective NSAIDs. It is characterized by a long half-life, allowing for convenient once-daily dosing. However, like other selective inhibitors, it carries a potential risk of cardiovascular side effects, such as hypertension and an increased risk of thrombotic events. Therefore, it is contraindicated in patients with established ischemic heart disease, peripheral arterial disease, or uncontrolled high blood pressure and is usually prescribed at the lowest effective dose for the shortest duration necessary.
