Indications:Diasulin is used for the treatment of type 1 diabetes and type 2 diabetes when not adequately controlled by diet or oral medications. It is also used in diabetic ketoacidosis, hyperosmolar states, during severe infections or surgery, and in gestational diabetes.Composition:Available as 30/70 (30% regular insulin + 70% isophane insulin) and 50/50 mixtures, each containing 100 IU/ml of human insulin (rDNA).Pharmacology:Insulin lowers blood glucose by increasing glucose uptake in muscle and fat cells and reducing glucose production in the liver. After subcutaneous injection, onset occurs within 30 minutes, peak effect is seen in 1–3 hours, and duration lasts about 18–24 hours.Dosage and Administration:Dosage is individualised based on patient needs. Typical daily insulin requirements range from 0.5–1.0 IU/kg in type 1 diabetes and 0.3–0.6 IU/kg in type 2 diabetes. It is given subcutaneously, usually in the abdomen (preferred), thigh, or arm. Injection should be followed by a meal within 30 minutes. Injection sites should be rotated to avoid lipodystrophy.Drug Interactions:Insulin requirements may decrease with oral antidiabetics, ACE inhibitors, MAO inhibitors, beta-blockers, salicylates and alcohol. Requirements may increase with corticosteroids, thyroid hormones, thiazides and sympathomimetics. Beta-blockers may mask hypoglycemia symptoms.Contraindications:Contraindicated in hypoglycemia and in patients with hypersensitivity to insulin or its components.Side Effects:The most common side effect is hypoglycemia. Other effects include injection site reactions, lipodystrophy, edema and allergic reactions, which may rarely be severe.Precautions and Warnings:Incorrect dosing or stopping insulin may lead to hyperglycemia or diabetic ketoacidosis. Dose adjustments may be required with changes in diet, activity, illness or insulin type. Do not use if the insulin appears abnormal after mixing.Pregnancy and Lactation:Insulin is safe in pregnancy and does not cross the placenta. Dose requirements may change during pregnancy and after delivery. It is also safe during breastfeeding, but dose and diet adjustments may be needed.Overdose Effects:Overdose may cause hypoglycemia. Mild cases can be treated with oral glucose, while severe cases require glucagon injection or intravenous glucose.Storage Conditions:Store in a refrigerator at 2°C–8°C. Do not freeze. Protect from light and heat. In-use cartridges can be kept below 25–30°C for a limited time. Mix well before use.⚠️ Use only under the supervision of a registered healthcare professional.
