Product Description

Indications: A-Flox is indicated for the treatment of infections caused by Gram-positive bacteria, including penicillinase-producing staphylococci. It is used in skin and soft tissue infections such as boils, abscesses, carbuncles, infected ulcers, eczema, acne, cellulitis, infected wounds, burns, impetigo, otitis media and externa. It is also indicated for respiratory tract infections including pneumonia, lung abscess, empyema, sinusitis, pharyngitis, tonsillitis and quinsy. Additionally, it is used in osteomyelitis, enteritis, endocarditis, urinary tract infections, meningitis and septicemia caused by susceptible organisms. It may also be used as a prophylactic agent during major surgical procedures such as cardiothoracic and orthopedic surgeries.Pharmacology: Flucloxacillin is a bactericidal isoxazolyl penicillin that is resistant to penicillinase and effective mainly against Gram-positive organisms. It acts by inhibiting bacterial cell wall synthesis through blocking transpeptidase enzyme activity, thereby interfering with peptidoglycan cross-linking. This weakens the bacterial cell wall, leading to cell lysis and death. Its structural side chain provides resistance to β-lactamase enzymes.Dosage & Administration: Oral administration: Adults are usually given 250 mg four times daily, which may be increased in severe infections; in conditions such as osteomyelitis and endocarditis, doses up to 8 g daily in divided doses may be required. Children aged 2–10 years receive half of the adult dose, and children under 2 years receive one-quarter of the adult dose. Parenteral administration: Adults may receive 250 mg four times daily by intramuscular injection or 250 mg to 1 g four times daily intravenously. In severe infections, total daily doses may be increased up to 8 g. Pediatric doses are adjusted similarly to oral dosing (half or one-quarter of adult dose depending on age).Administration: Oral doses should be taken at least 1 hour before meals.Interaction: Concurrent use with certain drugs may increase the plasma concentration of flucloxacillin and prolong its action.Contraindications: Contraindicated in patients with known hypersensitivity to flucloxacillin or other penicillins.Side Effects: Common adverse effects include gastrointestinal disturbances such as nausea, vomiting, diarrhea and dyspepsia. Other reported effects include rash, urticaria, purpura, fever, interstitial nephritis, hepatitis and cholestatic jaundice.Pregnancy & Lactation: Classified as pregnancy category B. It should be used during pregnancy only if clearly necessary. Flucloxacillin is excreted in breast milk; therefore, caution is advised in lactating mothers.Precautions & Warnings: Use with caution in patients with hepatic dysfunction or allergic conditions. Monitoring is advised in prolonged therapy.Use in Special Populations: In patients with severe renal impairment (creatinine clearance <10 ml/min), dose reduction or adjustment of dosing interval may be required.Therapeutic Class: Penicillinase-resistant penicillinsReconstitution: Flucloxacillin may be administered via intra-articular or intrapleural routes in specific cases. It may also be prepared for nebulization by dissolving 125–250 mg in sterile water for inhalation.Storage Conditions: Store in a dry place, protected from light and heat. Keep out of the reach of children.
