Product Description

Indications: Joinix D tablet is indicated for the treatment of osteoarthritis, rheumatoid arthritis, bone injuries, and joint injuries.


Pharmacology:


Glucosamine is a naturally occurring amino sugar that plays a key role in the formation and repair of cartilage. It is an essential precursor for the synthesis of glycosaminoglycans (GAGs) such as hyaluronic acid, keratan sulfate, and chondroitin sulfate, which combine with proteins to form proteoglycans — the major structural components of articular cartilage. Supplementation with glucosamine sulfate may help stimulate cartilage repair and support joint function by activating chondrocytes responsible for producing cartilage matrix components. Approximately 90% of orally administered glucosamine sulfate is absorbed through the gastrointestinal tract. It is mainly metabolized in the liver and excreted through urine.


Diacerein is used in the treatment of osteoarthritis and possesses anti-inflammatory, analgesic, and antipyretic properties. It acts by inhibiting Interleukin-1 (IL-1), a cytokine involved in cartilage degradation and joint inflammation. Due to its unique mechanism of action, Diacerein may provide disease-modifying effects in osteoarthritis, especially in knee and finger joints. Oral bioavailability is around 56%. Food delays peak concentration but increases absorption by approximately 25%, therefore it is recommended to take with meals. Diacerein is extensively metabolized in the liver and excreted mainly in urine as metabolites.


Dosage & Administration: Adults are usually advised to take one tablet twice daily with food. Safety and effectiveness in children and adolescents below 18 years of age have not been established.


Interaction: Aluminium and magnesium hydroxide-containing antacids may reduce the absorption of Diacerein. Concurrent use with laxatives or antibiotics may increase the risk of diarrhea.


Contraindications: Contraindicated in patients with known hypersensitivity to Glucosamine, Diacerein, or any component of the formulation.


Side Effects: Generally well tolerated. Reported side effects include nausea, vomiting, diarrhea, epigastric pain, headache, skin rash, and yellow discoloration of urine.


Pregnancy & Lactation: Contraindicated during pregnancy and breastfeeding.


Precautions & Warnings: Use cautiously in patients with known allergies to Glucosamine or Diacerein. Monitoring is advised if gastrointestinal side effects occur.


Overdose Effects: No specific data regarding overdose is currently available.


Therapeutic Class: Cartilage formation stimulants.
