Indomethacin is a highly potent, non-selective nonsteroidal anti-inflammatory drug (NSAID) belonging to the methylated indole class, recognized for its robust analgesic, antipyretic, and anti-inflammatory capabilities. It functions primarily by achieving powerful inhibition of the enzymes cyclooxygenase-1 and -2 (COX-1 and COX-2), which effectively halts the synthesis of prostaglandins from arachidonic acid. While it is used for managing moderate to severe osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis, it is particularly distinguished by its status as a first-line treatment for acute gouty arthritis and various forms of hemicrania.
